When a T cell recognizes an antigen, the TCR complex triggers a cascade of phosphorylation events. HPK1 is recruited to the signalosome and phosphorylates (at Serine 376). This phosphorylation event creates a docking site for the E3 ubiquitin ligase Itch , leading to the degradation of SLP-76 and ultimately dampening the activation of MAP kinases (ERK, JNK) and NF-κB.
